Abstract

• We have used the electron microscopic immunogold techniqueto localize precisely various extracellular matrix componentsin human retinal vessels and Bruch's membrane. Collagen typesIV and V, laminin, and heparan sulfate proteoglycan core proteinwere localized in basement membranes of retinal capillaries.In addition to the capillary basement membrane components, collagentypes I and III and fibronectin were found in the basement membranesof retinal arterioles and venules. The basement membranes ofchoriocapillaries and retinal pigment epithelial cells in Bruch'smembrane also showed a similar distribution of the retinal capillarybasement membrane components. Both the inner and outer collagenouslayers of Bruch's membrane contained collagen types I and IIIalong with fibronectin, whereas collagen type VI was mostlylimited to the central elastic lamina. The precise localizationand distribution of various extracellular matrix componentsin human retinal vessels and Bruch's membrane may be importantfor understanding their normal function as well as their alterationin disease.
